308. The boundary of sedimentation (the boundary between the cloudy and transparent parts of the solution) of horse hemoglobin (M = 68350 g/mol) in an aqueous solution at 30 0C was observed as a function of sedimentation time at a centrifuge speed of 39300 min-1:
t, min 45 75 105 135 165
x*, cm 4.525 4.601 4.679 4.757 4.84
* x - distance from the axis of rotation
Based on these data, determine the sedimentation coefficient s, the diffusion coefficient D, and the friction coefficient for this protein, if it is known that the specific partial volume of the protein is 0.755 cm3/g, the density of the solvent is 0.996 g/cm3. Calculate also the coefficient of friction of the equivalent sphere f0 using the Stokes equation and find the ratio f/fo. The viscosity of the solvent is 8.02*10-4 Pa*s.
